- VACCINS SYNTHETIQUES Lobjectif de ce projet est de dvelopper des vaccins synthtiques
prophylactiques ou thrapeutiques dirigs contre une bactrie, un virus ou une tumeur. Ce projet est
bas sur lutilisation de systmes nanoparticulaires, notamment de liposomes, comme vecteurs des
lments indispensables linduction dune rponse immunitaire forte et spcifique. Nous dveloppons
diffrentes techniques de chimie de bioconjugaison de ces lments au vecteur. Nous ralisons la
synthse (adjuvants et molcules de ciblage), la mise au point, la caractrisation et lvaluation in vitro
et in vivo des diffrents vecteurs nanoparticulaires formuls
- CHIMIE DES POLYMERES / BIOMATERIAUX Nous utilisons toutes les techniques de
bioconjugaison pour modifier chimiquement diffrents polymres dintrts biologiques (polylysine,
polyglutamique acide, chitosane, hyaluronane). Ces polymres seront utiliss pour construire des
films de polylectrolytes auto-assembls (par la technique de "layer-by-layer") qui vont permettre par
simple recouvrement, la modification de surfaces de biomatriaux (prothses de types osto-
articulaires, dentaires, vaisseaux artificiels), ceci afin de leur confrer des proprits nouvelles. Une
nouvelle technique de fabrication de biomatriaux ultracentrifugs en prsence de sel base de
polymres naturels est en cours dtude. Les premiers rsultats sont extrmement prometteurs tant du
point de vue mcanique, physico-chimique ou toxicologique. Ces nouveaux biomatriaux pourront tre
utiliss comme agent de dlivrance de principes actifs sous forme dimplant, de microparticules et
mme de nanoparticules.
- TOXICOCINETIQUE ET TOXICO-DYNAMIQUE DES NANOPARTICULES
MANUFACTUREES Les effets nocifs des particules ultrafines associes la pollution atmosphrique,
largement dcrits au cours de dernires annes, font craindre des effets indsirables des nanoparticules
manufactures sur la sant. Nos recherches ont pour objectif de contribuer llaboration de
nanoparticules optimises en termes de toxicit. Notre exprience en exprimentation animale est mise
profit pour tudier la biodistribution des nanoparticules marques chez la souris. Ces tudes de
biodistribution sont associes une valuation de la toxicit (mortalit, biochimie clinique des
principaux organes, activit immunomodulatrice ou immunogne)

- Lquipe explore les mcanismes molculaires dactivation de canaux ioniques activs par un ligand.
En particulier, elle sintresse aux rcepteurs P2X activs par lATP extracellulaire qui constituent une
famille mergente de canaux impliqus dans de grandes fonctions physiopathologiques comme les
douleurs neuropathiques. Elle cre galement de nouveaux outils optogntiques permettant dactiver
plus slectivement ces canaux ioniques par la lumire.
Le groupe de Chimie dveloppe des outils chimiques pour la photo-rgulation dactivits biologiques.
Deux stratgies sont envisages afin de photo-rguler une activit biologique :
- Soit en utilisant une raction de photo-isomrisation permettant de photo-rguler une interaction ligand-
rcepteur.
- Soit en utilisant une raction de photo-clivage permettant de librer de faon contrler un effecteur
biologique sous laction de la lumire. Lutilisation dune raction de photo-clivage est un concept pour
lequel le principal enjeu scientifique actuel rside dans lutilisation dune excitation 2-photons. Ce
type dexcitations permet de palier au problme de rsolution spatial quengendre lutilisation classique

dune excitation mono-photonique lors de la photolyse et permet de plus, denvisager une utilisation sur
des tissus ou des organisme vivant du faite dune plus grande pntration tissulaire.

ACTIVITES DE RECHERCHE
PROJET SCIENTIFIQUE GLOBAL DU LABORATOIRE
La connaissance du Vivant est une qute sans fin. La comprhension des mcanismes biologiques
lmentaires au niveau molculaire a fait des progrs considrables mais le champ dinvestigation est
encore immense. Il reste par ailleurs dsormais remonter de llmentaire au complexe selon une
dmarche que lon pourra vouloir rductionniste ou holistique. Il nous semble que notre modeste
contribution cette recherche sur le Vivant reste plus que jamais pertinente car nous souhaitons concevoir
et fournir les molcules-outils qui permettront dexplorer le Vivant dans tous ses niveaux de complexit,
de la molcule lanimal entier, jusqu lhomme. Aux cts des nouvelles techniques omiques
(gnomique, protomique, mtabolomique, etc.) il nous semble en effet crucial de pouvoir perturber et
analyser tout systme avec la souplesse quautorisent les sondes chimiques fonctionnelles. Nous garderons
donc comme projet de recherche le cap que nous nous sommes fixs depuis 15 ans, savoir :
Rechercher et dvelopper des petites molcules originales
permettant dtudier le Vivant et de le soigner.
Dans quels domaines ?
Toutes nos quipes contribuent concevoir et produire des molcules toujours plus diverses et originales,
soit synthtiques soit naturelles, afin dlargir la palette de substances disponibles pour perturber et
analyser le Vivant. Cest loccasion de faire progresser les stratgies et les mthodes de modlisation, de
synthse organique et dextraction. La diversit structurale engendre vient enrichir la chimiothque
nationale et europenne et la probabilit de trouver des touches originales sur des cibles de plus en plus
complexes.
Le bilan du quadriennal prcdent montre la force de notre position dans le domaine des Rcepteurs
coupls aux protines G et de leurs voies de signalisation (en particulier le calcium). Cet axe de recherche
est trs concurrentiel mais de nombreux processus fondamentaux restent explorer et exploiter et notre
avantage comptitif est significatif. Nous poursuivons donc nos travaux sur les projets les plus innovants
dans le domaine, tant en termes mcanistiques (structure, allostrie, oligomrisation fonctionnelle,
marquage) que thrapeutiques (cibles non exploites).
Nous concentrons nos efforts sur des axes thrapeutiques o les attentes socitales sont fortes et pour
lesquels nous avons un angle dattaque fort et original correspondant une prise de risque certaine mais
calcule. Il sagit :
1) du systme nerveux central avec deux axes forts : la douleur sous langle de lhyperalgsie ; lamour
sous son angle affect, avec des applications dans lautisme et la motivation socitale.

2) de linflammation, avec des concepts thrapeutiques originaux (neutraligands), des biomarqueurs
pertinents et un portfolio de cibles valides.
3) du cancer, avec ltude et linhibition spcifique de cellules souches tumorales.
4) des maladies rares ou ngliges, en combinant approche ethnopharmacologique et approche
thrapeutique classique.
Les axes Cancer et Inflammation sont au cur du Laboratoire dExcellence MEDALIS et ont t jugs
pertinents et comptitifs par son panel dexperts internationaux.
EQUIPE CHIMIE-BIOLOGIE INTEGRATIVE

1- Problmes scientifiques abords
Notre objectif est de dcouvrir rapidement des petites molcules permettant de comprendre le Vivant et de
le manipuler dans une perspective cognitive ou/et thrapeutique. Nous avons abord ces problmes selon
quatre angles correspondant quatre thmatiques :
Thmatique 1 : la Chemical Biology. Nous dveloppons de nouvelles voies de synthses autour
de Diversophores pour produire des sondes originales et potentiellement bioactives. Nous
concevons par ailleurs des sondes traables pour ltude mcanistique de RCPG et des
chimiothques traables adaptes la microfluidique pour acclrer et optimiser les tudes
dinteractomique complexe.
Thmatique 2 : Le cancer et linflammation. Nous recherchons les mcanismes daction et
poursuivons le dveloppement thrapeutique des molcules innovantes rcemment dcouvertes,
agissant comme neutraligands de chimiokines ou comme inhibiteurs de cellules souches
cancreuses. Nous cherchons en parallle des biomarqueurs cliniques et validons de nouvelles
cibles thrapeutiques.
Thmatique 3 : lamour, la douleur. Nous cherchons et dveloppons de nouveaux composs
agissant sur les rcepteurs vasopressine/ocytocine (Amour) et sur divers rcepteurs impliqus dans
la douleur et lhyperalgie.
Thmatique 4 : les maladies parasitaires. Notre objectif actuel est doptimiser par chimie
mdicinale les activits antipaludique et/ou leishmanicide de molcules naturelles identifies par
fractionnement bio-guid, didentifier les mcanismes daction ainsi que les cibles molculaires
impliqus.

SYNTHESE ORGANIQUE METALLOCATALYSEE ET PHARMACOCHIMIE
Responsable : Jean Suffert
Le laboratoire dveloppe aujourdhui des mthodologies synthtiques originales autour de la raction de
cyclocarbopalladation. La mise au point de processus synthtiques en cascade mens autour de cette
raction permet llaboration de systmes polycycliques particulirement complexes comme par exemple
les fenestradines et les cyclooctatrines. Lapplication de ces mthodologies dans le domaine de composs
incluant dans leurs structures un atome de soufre ou silicium est actuellement en cours et a dores et dj
permis laccs de nouvelles familles de molcules originales.
Notre quipe tudie galement laccs des molcules htrocycliques par des ractions domino
multicomposantes mdies au cuivre. Cette approche nous a notamment permis de dvelopper une synthse
originale daminobenzothiazoles polyfonctionnaliss.
Une autre partie de lquipe s'intresse l'obtention de chssis molculaires complexes, souvent inspirs par
les substances naturelles, par lintgration de la raction d'hydroformylation dans des processus synthtiques
en cascade.
De plus, une thmatique axe sur la pharmacochimie nous a conduit participer la conception de
nouvelles plateformes molculaires anticancreuses et anxiolytiques.
EQUIPE CHEMOGENOMIQUE STRUCTURALE
Responsable : D. Rognan
Notre quipe se situe linterface de diverses disciplines (Chimie, Biologie, Informatique) et a pour but de
rationaliser et dacclrer lidentification de molcules bioactives dans deux buts bien prcis :
fournir des sondes biochimiques et pharmacologiques permettant dtudier une protine cible et
son implication dans des contextes pathologiques,

proposer des touches au chimiste mdicinal afin den amliorer les proprits (efficacit,
slectivit).
Pour ce faire, nous dveloppons et appliquons de nouveaux algorithmes en cheminformatique et
bioinformatique afin de mieux dcrire lespace chimique des ligands, lespace biologique des cibles et
lespace protine-ligand.

Le domaine dexpertise de lquipe concerne les substances naturelles obtenues par phytochimie
extractive, et dotes de proprits nouvellement dcrites ou relevant dun mcanisme daction original.
Notre objectif est didentifier et de valoriser ces substances en mettant profit la chimiodiversit
molculaire puise dans le rgne vgtal.
Les stratgies mises en place sappuient la fois sur notre expertise interne en Pharmacognosie et sur la
consolidation de nos rseaux de collaborations en botanique-chimie et biologie.

- Profiling dextraits complexes puis stratgie de drplication en vue dacclrer lisolement de nouvelles

Plantes halophiles
Lobjectif poursuivi est didentifier les principaux facteurs susceptibles dimpacter le mtabolisme
secondaire despces vgtales croissant en bord de littoral et soumises des stress hydrique et salin. Nous
comparons galement les capacits biosynthtiques de plantes sauvages, de plantes cultives et de cellules
souches vgtales produites en biofermenteurs.
Nos rsultats nous permettent dassurer une slection rationnelle des espces dintrt et de les valoriser
tout en prservant leur biotope naturel.

En associant plusieurs techniques phytochimiques et chromatographiques, notamment grce au couplage
LC-MS-SPE-RMN et un prfractionnement par CPC, et grce une stratgie de drplication, nous
avons explor la composition chimique de deux orchides de la sous-tribu des Aeridinae. Dix drivs
phnanthrniques ont t identifis dans les tiges dAerides rosea Lodd. ex. Lindl. & Paxton, dont deux
nouvellement dcrits.Quatre stilbnodes, trois drivs d'acides phnoliques et quatre esters d'acide
cinnamique ont t identifis dans les tiges dAcampe rigida (Buch.-Ham. ex Sm.) P. F. Hunt.
Nous avons galement effectu des dosages de traceurs dans diffrents chantillons de deux reprsentants
de la tribu des Vandeae : Vanda teres (Roxb.) Lindl. et Vanda coerulea Griff. ex. Lindl., afin de mettre en
vidence dventuelles variations de composition en fonction des facteurs environnementaux et du stade
de croissance vgtative. Nous avons ainsi compar par LC-MS-MS les teneurs en marqueurs dactivit de
divers chantillons variant selon leur origine (rserve naturelle en Chine, ferme en Thalande, serre en
Suisse), lge des plantes, leur priode de prlvement (avant, pendant et aprs floraison) et la nature des
organes slectionns (tiges, feuilles, racine).
Enfin, nous avons mis au point des conditions disolement prparatif par CPC afin de purifier des
marqueurs biologiques pralablement identifis dans les tiges de Vanda teres : trois
glucospyranosyloxybenzyl-malates ainsi que leur prcurseur biosynthtique.
B. Scrtagogues naturels du GLP-1 (resp. A. Urbain)
L'un des projets de l'quipe consiste rechercher de nouveaux candidats mdicaments pour le traitement
du diabte de type 2, et plus prcisment des scrtagogues du GLP-1 (glucagon-like peptide-1). En effet,
la scrtion du GLP-1, hormone implique dans la rgulation de la glycmie via notamment la production
d'insuline, est diminue chez les diabtiques de type 2.
Lors d'un criblage cellulaire in vitro, une stimulation de la scrtion du GLP-1 a t observe avec des
extraits issus dApocynaces malgaches et dAsteraces polynsiennes, aucune de ces espces n'ayant fait
lobjet jusqu' prsent dtudes phytochimique et pharmacologique. Le fractionnement bioguid des
extraits bioactifs est en cours afin d'identifier la ou les molcule(s) responsable(s) de l'activit observe.
L'identification des principes actifs se fera notamment l'aide du couplage HPLC-UV-MS/MS-RMN
capillaire.
C. Substances naturelles pro-apoptotiques (resp. CD. Muller)
De nouveaux outils pharmacologiques capables dactiver lapoptose sur diffrents modles cellulaires ont
t gnrs partir dextraits naturels dans le cadre dun programme europen (FP7- MAREX :
"Exploring Marine Resources for Bioactive Compounds : From Discovery to Sustainable Production and
Industrial Application ). Ce programme nous a donn accs une importante chimiothque de molcules
naturelles issues dorganismes marins et constituant des sources potentielles de produits haute valeur
ajoute. Des extraits ont t tests pour leurs activits biologiques : anticancreuses, anti-inflammatoires,
antivirales et anti-coagulantes. A Strasbourg, nous avons apport notre expertise dans le criblage
anticancreux grce des mthodologies originales en cytomique, utilisant 3 lignes cancreuses humaine
(testiculaire, hpatique et sanguine) ainsi quune ligne cutane non cancreuse. L'isolement des
composs bio-actifs suivi par la dtermination de leurs structures a permis aux pharmaco-chimistes
slovnes (Pr. Danijel Kikelj, Universit de Ljubljana) impliqus dans ce programme europen, de
produire de nouveaux analogues synthtiques bioactifs, capable dinduire lapoptose par la voie
extrinsque en activant la caspase-8.
