hydroxychloroquine (n=414) (800 mg once, followed by 600 mg in 6 to 8 hours, then 600 mg daily for 4 days).
• Incidence of new illness compatible with Covid-19 not significant between
hydroxychloroquine (49 of 414 [11.8%]) and placebo (58 of 407 [14.3%]); absolute
difference −2.4% (95% confidence interval, −7.0 to 2.2; P=0.35).
• Side effects were more common with hydroxychloroquine than with placebo (40.1% vs. 16.8%), but no serious adverse reactions were reported.

Randomised, double-blind, placebo-controlled, multicentre trial at ten hospitals in Hubei, China.
Eligible patients: adults hospitalized with COVID-19 and oxygen saturation of 94% or less on room air
237 patients enrolled and randomly assigned to a treatment group (158 to remdesivir and 79 to placebo)
Primary endpoint = time to clinical improvement up to day 28, defined as the time (days) fromrandomisation to two levels decline on six-point ordinal scale of clinical status
Early termination because of the lack of new patients, with the actual enrolment of 236 participants, statisticalpower was thus reduced from 80% to 58%
Remdesivir use was not associated with a difference in time to clinical improvement (median remdesivir21·0 days [IQR 13·0–28·0] vs placebo 23·0 days [15·0–28·0]; HR 1·23 [95% CI 0·87–1·75])
Although not statistically significant, patients receiving remdesivir had a numerically faster time to clinicalimprovement than those receiving placebo among patients with symptom duration of 10 days or less: median18·0 days[IQR 12·0–28·0]vs 23·0 days [15·0–28·0]; HR 1·52 [0·95–2·43]
28-day mortality was similar between the two groups (22 [14%] died in the remdesivir group vs 10 (13%) in the placebo group; difference 1·1% [95% CI –8·1 to 10·3])

Double-blind, randomized, multicentre placebo-controlled trial mainly in the USA
Adults hospitalized with Covid-19 and evidence of lower respiratory tract involvement
538 patients assigned to remdesivir and 521 to placebo
Primary outcome = the time to recovery, defined by either discharge from the hospital or hospitalization for infection-control purposes only
The data and safety monitoring board recommended early unblinding of the results on the basis of findings from an analysis that showed shortened time to recovery in the remdesivirgroup
Patients who received remdesivir had a median recovery time of 11 days (95% confidence interval [CI], 9 to 12), as compared with 15 days (95% CI, 13 to 19) in those who received placebo (rate ratio for recovery, 1.32; 95% CI, 1.12 to 1.55; P<0.001)
The Kaplan-Meier estimates of mortality by 14 days were 7.1% with remdesivir and 11.9% with placebo (hazard ratio for death, 0.70; 95% CI, 0.47 to 1.04).
